Scientific Background :HER3 is a member of the human epidermal growth factor receptor (EGFR/HER) tyrosine kinase family whose kinase domain lacks several residues critical for catalysis but serves as an activator of the EGFR kinase domain (1,2). Phosphorylation of HER3 by other EGFR family members, most commonly HER2, activates the PI3K/AKT pathway (2). In cancer cells treated with HER2 kinase inhibitors, diminished HER2 activity is compensated for by HER3 overexpression, resulting in tumor cell survival (2).
