Scientific Background : CYLD is a cytoplasmic deubiquitinating enzyme, which removes ubiquitin molecules from various signaling proteins, and regulates the activities of many cellular and signaling processes. It plays a role in the regulation of inflammation, immunity, cell cycle progression, spermatogenesis, osteoclastogenesis, ciliogenesis, migration and tumorigenesis. (1) Several signaling pathways, such as the Nuclear Factor-kB (NF-kB), Wnt/β-catenin and c-Jun N-terminal kinase (JNK) pathways, and potentially others have been linked with or regulated by CYLD. (2) Mutations in CYLD gene have been associated with cylindromatosis, multiple familial trichoepithelioma, and Brooke-Spiegler syndrome. (3)
