Scientific Background :Thrombin is an endolytic serine protease that cleaves fibrinogen to produce insoluble active fibrin strands and fibrinopeptides A and B. This enzymatic process is essential for blood clot formation. Thrombin recognizes the consensus sequence A-B-Pro-Arg-▼-X-Y, with the first two amino acids representing hydrophobic residues and the last two amino acids representing non-acidic residues. The common recognized sequences are Leu-Val-Pro-Arg-▼-Gly-Ser and Gly-Arg-▼-Gly. Thrombin has high proteolytic specificity and hydrolytic efficiency, making it a widely used tool for cleaving purification tag  from recombinant fusion proteins.
