Scientific Background :Recombinant Tobacco Etch Virus (TEV) Protease is derived from the naturally occurring 28 kDa Nla recombinant protease. TEV Protease is commonly used for removing affinity tags from purified recombinant fusion proteins. TEV protease is highly site-specific, targeting the consensus sequence EXXYXQ(G/S). Cleavage occurs between Q and G/S. The most common TEV target sequences are ENLYFQG or ENLYFQS. The enzyme retains activity in solutions containing 1M Urea, 1-5mM DTT, beta-Mercaptoethanol, EDTA and PMSF, Aprotinin, Leupeptin and other protease inhibitors. The 6-His tag enables easy removal of the protease.
