Expression System:

Sequence :

Specificity :Recognizes the p70S6K protein

Cited Applications :WB

Cross Reactivity :Western blot p70S6K from human and mouse cells

p70S6K from other species may also be detectable.

Host  :Rabbit

Immunogen :Synthetic peptide corresponding to amino acids 409-413 of human p70S6K (protein accession number P23443)

Purification :Affinity chromatography

Stability :Store at 4oC (add 0.1% NaN3) for several months, and at -20oC for longer periods. For optimal storage, aliquot target into smaller quantities after centrifugation and store at recommended temperature. For optimal performance, avoid repeated handling and multiple freeze/thaw cycles.

Sample Data :Representative western blot with Anti-p70S6K (1:1000) using 5 ng of activated His-tag human recombinant p70S6K.

Sample Data :Representative western blot with Anti-p70S6K (1:1000) using 30 μg of HeLa, HEK-293T, Jurkat, THP1, and 3T6 cell lysate.

Scientific Background :p70S6K is responsible for the phosphorylation of 40S ribosomal protein S6, and is ubiquitously expressed in human adult tissues (1). p70S6K is activated by serum stimulation and this activation is inhibited by wortmannin and rapamycin. p70S6k activity changes during the cell cycle, and increases 20-fold in G1 cells released from G0 (2). p70S6K activation requires sequential phosphorylation at proline-directed residues in the putative autoinhibitory pseudosubstrate domain, as well as threonine 389, a site phosphorylated by phosphoinositide-dependent kinase 1 (PDK-1).
